Concentrations of polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), polychlorinated biphenyls (PCBs), and organochlorine pesticides were measured in the serum of a sample of the New Zealand population aged 15 years and older. This was the first study to obtain representative measures of PCDDs, PCDFs and PCBs in the adult population of an entire country. Serum samples were obtained in 1996-1997. Potentially occupationally exposed individuals were excluded. Serum samples were pooled according to stratification criteria for area of residence, ethnicity, age, and sex. Of the 80 possible strata, sufficient serum for chemical analysis was available for 60, to which 1,834 individual samples contributed. For the PCDDs and PCDFs, most 2,3,7,8-chlorinated congeners were measured in all strata, with a mean toxic equivalents concentration across all strata of 12.8 ng TEQ kg(-1) lipid. Seven PCB congeners were frequently measured, including the coplanar congeners #126 and #169, quantified in all strata. Of the pesticides and their metabolites, only beta-HCH, dieldrin and pp'-DDE were consistently detected across strata. There was a general trend of increasing concentration with age. There were no consistent differences between the sexes, or between people of Maori (the indigenous people of New Zealand) and non-Maori ethnicity. Concentrations of PCDDs and PCDFs tended to increase in a North-South direction, possibly reflecting greater levels of industrialization and population concentration, and concentrations of the pesticide products were highest in the South, possibly reflecting historical use patterns. Results were consistent with a recent study of concentrations of these compounds in the milk of first-time mothers.  相似文献

There are many challenges in the accurate quantification of bacterial genes, such as the atrazine-degrading enzyme antA from Pseudomonas sp. strain ADP, from soil samples. We compared four quantitative methods for enumeration of atrazine-degrading bacteria in rhizosphere environments and utilized the optimal probe-based real-time polymerase chain reaction (PCR)-based method in an ongoing bioremediation experiment to monitor atzA copy number over time. We compared three quantitative PCR (qPCR) based methods--quantitative competitive PCR and two real-time qPCR methods--to traditional dilution-plate counting techniques. The optimal real-time qPCR assay was then used to monitor atzA copy number over time in the robust atrazine-degrading Pseudomonas sp. strain ADP-spiked rhizosphere environment. The use of sensitive and reliable probe-based real-time qPCRs for the enumeration of bacterial catabolic genes allows for their detection from soil samples and monitoring of potential degradative populations over time. The addition of arrazine-biodegrading bacteria into arrazine-contaminated sites to remove entrapped atrazine is a promising approach for mitigating atrazine pollution and its metabolites. The methodology contained herein will allow for optimal monitoring of atzA in rhizosphere soil with or without the addition of biodegradative Pseudomonas sp. strain ADP of bacteria.  相似文献

A sound multi-species vegetation buffer design should incorporate the species that facilitate rapid degradation and sequestration of deposited herbicides in the buffer. A field lysimeter study with six different ground covers (bare ground, orchardgrass, tall fescue, timothy, smooth bromegrass, and switchgrass) was established to assess the bioremediation capacity of five forage species to enhance atrazine (ATR) dissipation in the environment via plant uptake and degradation and detoxification in the rhizosphere. Results suggested that the majority of the applied ATR remained in the soil and only a relatively small fraction of herbicide leached to leachates (<15%) or was taken up by plants (<4%). Biological degradation or chemical hydroxylation of soil ATR was enhanced by 20 to 45% in forage treatment compared with the control. Of the ATR residues remaining in soil, switchgrass degraded more than 80% to less toxic metabolites, with 47% of these residues converted to the less mobile hydroxylated metabolites 25 d after application. The strong correlation between the degradation of N-dealkylated ATR metabolites and the increased microbial biomass carbon in forage treatments suggested that enhanced biological degradation in the rhizosphere was facilitated by the forages. Hydroxylated ATR degradation products were the predominant ATR metabolites in the tissues of switchgrass and tall fescue. In contrast, the N-dealkylated metabolites were the major degradation products found in the other cool-season species. The difference in metabolite patterns between the warm- and cool-season species demonstrated their contrasting detoxification mechanisms, which also related to their tolerance to ATR exposure. Based on this study, switchgrass is recommended for use in riparian buffers designed to reduce ATR toxicity and mobility due to its high tolerance and strong degradation capacity.  相似文献

Antibiotics are routinely used in intensive animal agriculture operations collectively known as Concentrated Animal Feed Operations(CAFO) which include dairy, poultry and swine farms. Wastewater generated by CAFOs often contains low levels of antibiotics and is typically managed in an anaerobic lagoon. The objective of this research is to investigate the uptake and fate of aqueous sulfamethazine(SMN) antibiotic by alfalfa(Medicago sativa)grass grown under hydroponic conditions. Uptake studies were conducted using hydroponically grown alfalfa in a commercially available nutrient solution supplemented with 10 mg/L of SMN antibiotic. Analysis of alfalfa sap, root zone, middle one-third, and top portion of the foliage showed varying uptake rate and translocation of SMN. The highest average amount of SMN(8.58 μg/kg) was detected in the root zone, followed by the top portion(1.89 μg/kg), middle one-third(1.30 μg/kg), and sap(0.38 μg/kg) samples, indicating a clear distribution of SMN within the sampled regions. The ultraviolet(UV) spectra of parent SMN and translocated SMN identified in different parts of the plant present the possibility of metabolization during the uptake process. Uptake of SMN using alfalfa grown under hydroponic conditions has potential as a promising remediation technology for removal of similar antibiotics from wastewater lagoons.  相似文献

The U.S. Geological Survey's New Jersey and Iowa Water Science Centers deployed ultraviolet‐visible spectrophotometric sensors at water‐quality monitoring sites on the Passaic and Pompton Rivers at Two Bridges, New Jersey, on Toms River at Toms River, New Jersey, and on the North Raccoon River near Jefferson, Iowa to continuously measure in‐stream nitrate plus nitrite as nitrogen (NO3 + NO2) concentrations in conjunction with continuous stream flow measurements. Statistical analysis of NO3 + NO2 vs. stream discharge during storm events found statistically significant links between land use types and sampling site with the normalized area and rotational direction of NO3 + NO2‐stream discharge (N‐Q) hysteresis patterns. Statistically significant relations were also found between the normalized area of a hysteresis pattern and several flow parameters as well as the normalized area adjusted for rotational direction and minimum NO3 + NO2 concentrations. The mean normalized hysteresis area for forested land use was smaller than that of urban and agricultural land uses. The hysteresis rotational direction of the agricultural land use was opposite of that of the urban and undeveloped land uses. An r2 of 0.81 for the relation between the minimum normalized NO3 + NO2 concentration during a storm vs. the normalized NO3 + NO2 concentration at peak flow suggested that dilution was the dominant process controlling NO3 + NO2 concentrations over the course of most storm events.  相似文献

Conservation planners are faced with numerous choices regarding ‘what to connect’ when modeling landscape networks on an ecoregion scale. A simplifying assumption is often that coarse-scale corridors may provide overlapping or ‘umbrella’ effects for multiple conservation scenarios. To examine this, we assessed differences in connectivity models arising from four different conservation scenarios for the transboundary Northern Appalachian/Acadian Ecoregion (330,000 km2). Models resulted in networks comprised of varying levels of connectedness, suggesting the presence of local connectivity at several sub-ecoregion scales, but diminished ecoregion-scale connectivity. Our results also indicate that selecting ‘what to connect’ at the ecoregion scale strongly influences the location and extent of modeled corridors. Further, our evidence suggests that landscape networks derived for one scenario are not likely to produce far-reaching corridor umbrellas or highly coincident landscape networks for alternative conservation scenarios with varying goals. As a result, alternative landscape networks should not be considered functionally equivalent.  相似文献

A paired watershed study consisting of agroforestry (trees plus grass buffer strips), contour strips (grass buffer strips), and control treatments with a corn (Zea mays L.)-soybean [Glycine max (L.) Merr.] rotation was used to examine treatment effects on runoff, sediment, and nutrient losses. During the (1991-1997) calibration and subsequent three-year treatment periods, runoff was measured in 0.91- and 1.37-m H-flumes with bubbler flow meters. Composite samples were analyzed for sediment, total phosphorus (TP), total nitrogen (TN), nitrate, and ammonium. Calibration equations developed to predict runoff, sediment, and nutrients losses explained 66 to 97% of the variability between treatment watersheds. The contour strip and agroforestry treatments reduced runoff by 10 and 1% during the treatment period. In both treatments, most runoff reductions occurred in the second and third years after treatment establishment. The contour strip treatment reduced erosion by 19% in 1999, while erosion in the agroforestry treatment exceeded the predicted loss. Treatments reduced TP loss by 8 and 17% on contour strip and agroforestry watersheds. Treatments did not result in reductions in TN during the first two years of the treatment period. The contour strip and agroforestry treatments reduced TN loss by 21 and 20%, respectively, during a large precipitation event in the third year. During the third year of treatments, nitrate N loss was reduced 24 and 37% by contour strip and agroforestry treatments. Contour strip and agroforestry management practices effectively reduced nonpoint-source pollution in runoff from a corn-soybean rotation in the clay pan soils of northeastern Missouri.  相似文献

ABSTRACT: Both color infrared aerial photography and Landsat data are being used to provide information to meet present and future management goals for the Great Dismal Swamp National Wildlife Refuge. High and low altitude color infrared photographs are being used to study the hydrology and map the present vegetation of the swamp. A variety of significant ecologic units have been identified using these photographs. The completed maps will be used to evaluate analyses of landsat digital data. Once the present data base is compiled, it is hoped that routine analysis of Landsat data can be used for updating or to indicate areas where low altitude coverage or ground checking is desirable. The data base will also aid in identifying and evaluating trends that may provide guidelines for wetland management.  相似文献

ABSTRACT: A weighted average method was used to analyze transition zone vegetation in the Great Dismal Swamp to determine if a more uniform determination of wetland boundaries can be made nationwide. The method was applied to vegetation data collected on four transects and three vertical layers across the wetland-to-upland transition zone of the swamp. Ecological index values based on water tolerance were either taken from the literature or derived from local species tolerances. Wetland index values were calculated for 25-m increments using species cover and rankings based on the ecological indices. Wetland index values were used to designate increments as either wetland, transitional, or upland, and to examine the usefulness of a provisional wetland-upland break-point. Most increments were designated wetland or transitional when all species were used. Removal of three or five ubiquitous species either gave a wider range of wetland index values with a more variable designation of increments or caused designation of increments to be similar for all layers. The use of locally-derived rankings showed the sensitivity of the weighted averages method to ecological indices of species with large importance values. The weighted average method did not provide for an objective placement of an absolute wetland boundary, but did serve to focus attention on the transitional boundary zone where supplementary information is necessary to select a wetland-upland breakpoint.  相似文献
